Cytokine and Gene Expression Profiling in Patients with HFE-Associated Hereditary Hemochromatosis according to Genetic Profile
BACKGROUND: The hemochromatosis gene (HFE)-Hemochromatosis Hemochromatosis (HH) is characterized by regulating the synthesis of hepcidine, resulting in increased intake of intestinal iron.
Objectives: The objectives were to characterize and elucidate a possible association between the gene expression profile, the levels of hepcidin, the severity of the diseases and the inflammation markers of HH associated with HFE.
Methods: Thirty-nine HH HH patients HFE have been recruited and assigned to 2 groups according to the genetic profile: C282Y homozygous in a group and patients with H63D, such as homozygous or in association with C282Y, in the other group. Eleven healthy blood donors have been recruited as controls. Gene expression was characterized by peripheral blood cells and inflammatory cytokines and hepcidine-25 isoformation were quantified in serum. The characteristics of the biochemical disease have been recorded.
Results: High levels of Interleukin 8 have been observed in a greater proportion of patients than controls. In addition, compared to controls, ζ-globine gene expression has been considerably increased in homozygous C282y patients, while the expression of the matrix of metalloproteinase 8, and other proteins secreted by neutrophils, was significantly regulated in patients with H63D.
Conclusion: Different disease signatures can characterize HH patients depending on their HFE genetic profile. Studies on larger populations, including protein-level analyzes, are needed to confirm these results.
