Cytokine and Gene Expression Profiling in Patients with HFE-Associated Hereditary Hemochromatosis according to Genetic Profile

BACKGROUND: The hemochromatosis gene (HFE)-Hemochromatosis Hemochromatosis (HH) is characterized by regulating the synthesis of hepcidine, resulting in increased intake of intestinal iron.

Objectives: The objectives were to characterize and elucidate a possible association between the gene expression profile, the levels of hepcidin, the severity of the diseases and the inflammation markers of HH associated with HFE.

Methods: Thirty-nine HH HH patients HFE have been recruited and assigned to 2 groups according to the genetic profile: C282Y homozygous in a group and patients with H63D, such as homozygous or in association with C282Y, in the other group. Eleven healthy blood donors have been recruited as controls. Gene expression was characterized by peripheral blood cells and inflammatory cytokines and hepcidine-25 isoformation were quantified in serum. The characteristics of the biochemical disease have been recorded.

Results: High levels of Interleukin 8 have been observed in a greater proportion of patients than controls. In addition, compared to controls, ζ-globine gene expression has been considerably increased in homozygous C282y patients, while the expression of the matrix of metalloproteinase 8, and other proteins secreted by neutrophils, was significantly regulated in patients with H63D.

Conclusion: Different disease signatures can characterize HH patients depending on their HFE genetic profile. Studies on larger populations, including protein-level analyzes, are needed to confirm these results. To make the promise of CASP-CAS9-based genetics, approaches are necessary to quantify a molecular phenotype specific to all genetic libraries of genetic disturbances. We discussed this challenge by profiling transcription, translational and post-procedures journalists using crisis interference (CRISPRI) with the sequencing of the journalist of the bar expression (CIBER-SEQ). We found that the regulated genes at a time preferentially contained NF-Y binding patterns in their proximal promoters, including enriched genes related to ER functions rather than cell cycle.

This contrasts with the profiling data of the Hela and embryonic stem cells in which a separate regulation of cell cycle-related genes was observed. The grouping analysis has also identified several functional clusters where populations of low-pressure regulated genes were very distinct. Other analyzes using the immunoprecipitation of the chromatin and the DNA-SEQ data revealed that the transcriptomic difference was not correlated with the NF-Y DNA link but with NF-YA splice. These data suggest that neuronal cells have a different type of transcriptome wherein the ER-related genes are dominibly modulated by NF-Y and imply that the modification of the NF-YA splice may be involved in this gene modulation specific to the specific gene. cell.

In our study, 67 members of LEA (BDLEA) were identified in the Brachypodium Distachyon L genome. The analyzes of genes structure, evolutionary relations and protein reasons showed that the BDLEAS belonged to six sub-reflamal. The analyzes of the chromosomal locations and duplication numbers revealed that the 67 BDLeas were distributed on the five chromosomes and 26 BDLeas were identified as duplication event products. The results of the annotation of the ontology of genes (GO) suggested that nearly 60% of the BDLeas could be involved in the response to stress.
